It also accommodates advances in hardware and software.''Flying Objective'' technology increases scan speed, decreases optical distortion, and ensures emission path alignment by moving scanning mirrors and objective lens simultaneously. Optical path length and alignment are independent of scan-arm rotation, allowing uniform performance across the scan field throughout the Y-axis translation, and optimizing excitation/emission path lengths and optical alignments over the field of view. Aberrations and imperfections of earlier technology no longer dominate, improving scan linearity and gridding performance. Small scan arm can be positioned within a millimeter of the probe array. Large NA (0.62) objective is four times more efficient, allowing line scan speeds to easily reach more than fifty lines per second. Solid state 532nm diode-pumped, frequency doubled Nd:YAG (Neodymium-doped Yttrium Aluminum Garnet) Green Laser requires neither external power supply nor special cooling system. Optics condition the laser beam to deliver excitation spot size of 3.5um (two-fold improvement over the previous scanner). Significantly increases dynamic range (largest data value/smallest increment into which the data can be resolved or digitized). Can resolve 65,535 different levels of fluorescence (31 percent improvement in data precision over the previous scanner). Measurement noise is below the expected biological noise-floor of the array itself (a reduction over the previous scanner). Provides typical instrumentation system offset performance of around 20 counts, compensating for long-term offset effects in the scanner hardware.Immediately prior to each scan, shutters the photomultiplier tube to produce a ''dark'' scan (cumulative result of all electronic noise and offsets in the system at that time) and samples excitation laser power to compensate for changes in laser intensity from ambient temperature, dust, aging, etc.
